چکیده

Aortic aneurysm and dissections account for 1% to 2% of all deaths in the Western countries, and the incidence of thoracic aortic aneurysm (TAA) is ≈10.4 per 100 000 personyears. Furthermore, because acute aortic dissections might be disguised as heart attacks, true incidence numbers could be significantly higher. Aortic aneurysms are categorized into 2 main groups depending on their location: TAAs and abdominal aortic aneurysms. Abdominal aortic aneurysms are the most common group and are linked to various lifestyle-associated risk factors, including hyperlipidemia, hypertension, sex, and age. However, smoking is generally regarded as the most important risk factor. Heritability estimates as high as 70% have been found, and recently, several genes and loci have been associated with abdominal aortic aneurysms (DAB2IP, LRP1, CDKN2B-AS1, CNTN3, LPA, IL6R, and the sortilin-1 [SORT1] locus). In this review, we focus on TAA, which have an even stronger genetic component, with 20% of patients having a positive family history. This number is also likely to be underestimated because developing TAA usually remain asymptomatic. TAA can develop at the ascending Review

Genetics of thoracic aortic aneurysm: at the crossroad of transforming growth factor-β signaling and vascular smooth muscle cell contractility.

Aortic aneurysm, including both abdominal aortic aneurysm and thoracic aortic aneurysm, is the cause of death of 1% to 2% of the Western population. This review focuses only on thoracic aortic aneurysms and dissections. During the past decade, the genetic contribution to the pathogenesis of thoracic aortic aneurysms and dissections has revealed perturbed extracellular matrix signaling cascade i...

AMP-activated protein kinase inhibits transforming growth factor-β-mediated vascular smooth muscle cell growth: implications for a Smad-3-dependent mechanism.

Dysfunctional vascular growth is a major contributor to cardiovascular disease, the leading cause of morbidity and mortality worldwide. Growth factor-induced activation of vascular smooth muscle cells (VSMCs) results in a phenotypic switch from a quiescent, contractile state to a proliferative state foundational to vessel pathology. Transforming growth factor-β (TGF-β) is a multifunctional sign...

TGFβ Signaling-mediated MicroRNA Regulation in Vascular Smooth Muscle Cells

The discovery of small noncoding microRNAs (miRNAs) increases the complexity of gene expression regulatory mechanisms. The basic mechanism of miRNA biogenesis and regulatory functions of target genes have been widely elucidated. Recently, it has been recognized that regulation of each step of miRNA biogenesis is critical for generating functional miRNAs. Interestingly, cell signaling pathways, ...

Pharmacologically Improved Contractility Protects Against Aortic Dissection in Mice With Disrupted Transforming Growth Factor-β Signaling Despite Compromised Extracellular Matrix Properties.

OBJECTIVE Transforming growth factor-beta is a pleiotropic cytokine having diverse roles in vascular morphogenesis, homeostasis, and pathogenesis. Altered activity of and signaling through transforming growth factor-beta has been implicated in thoracic aortic aneurysms and dissections, conditions characterized by a reduced structural integrity of the wall that associates with altered biomechani...

Reactive Oxygen Species and p38MAPK Have a Role in the Smad2 Linker Region Phosphorylation Induced by TGF-β

Background: Transforming growth factor-β (TGF-β) in addition to the C-terminal region can phosphorylate receptor-regulated Smads (R-Smads) in their linker region. The aim of the present study was to evaluate the role of signaling mediators such as NAD(P)H oxidases (reactive oxygen species [ROS] generators), ROS, and ROS-sensitive p38 mitogen-activated protein kinase (p38MAPK) in this signaling ...
